Abstract

This talk will trace the evolution of a technique that has been dubbed In Situ Enzymatic Screening (ISES) wherein enzymes act as analytical tools/real-time sensors to assist synthetic chemists in the discovery of new catalytic reaction manifolds of interest and in the identification of useful chiral ligand families. For reactions for which this screening strategy is well suited, one obtains real time information on relative rates and/or enantioselectivity for an array of catalysts being screened in parallel, without the need to draw aliquots or quench the reactions. The nature of the signal readout and information content, as well as colorimetric and miniaturized versions of such bio-analytical screens will be discussed.

 

The application of ISES to the discovery of (i) the first examples of asymmetric allylic amination mediated by Ni(0)-catalysts; the identification of new (ii) bromorhodiation-carbocyclization and (iii) thiocyanopalladationcarbocyclization transformations, as well as (iv) the identification of useful new chiral salen ligands will be presented.

 

Biography

Dr. Berkowitz received his bachelor degree in University of Chicago in 1986. Then he became a traveling scholar in Eidgenössische Technische Hochschule (ETH) Zürich and did his doctorate in Harvard University until 1990. Then he joined in Prof. Samuel J. Danishefsky’s group as a postdoctoral in Yale University. In 1991, he worked as Assistant Professor of Chemistry in University of Nebraska. Then he became Associate Professor of Chemistry from 1997 to 2004 and worked as Professor of Chemistry from 2005 to 2010 in the department of chemistry of University of Nebraska. In 2011, he was awarded as Willa Cather Professor in the department of chemistry of University of Nebraska. He also works as Division Director in division of chemistry of National Science Foundation. In 2015, He was awarded as American Association for the Advancement of Science Fellow. And he became Visiting Professor in Universite de Paris-Descartes in 2016. Until now, he has been the author of many publications/patents and invited lecturer in conferences and universities.
